Design Notes

This design is codon optimized for expression in escherichia coli. We had some difficulty with synthesis of the repeats in the linker sequence and repeats in the BBa_B0034 terminator. The manganese peroxidase comes from “Pleurotus ostreatus manganese-dependent peroxidase silencing impairs decolourization of Orange II.” Salame et al. Microbiology and Biotechnology. 2010. GenBank: FJ594281.1 and the ligninase comes from “Nucleotide Sequence of a ligninase gene from Phanerochaete Chrsosporium.” Smith et al. Nucleic Acids Research. Volume 16. 1988.

This sequence was optimized for reducing repeats, palindromic sequences and restriction enzyme sites.
